没食子儿茶素没食子酸酯降低P糖蛋白表达逆转人耐药鼻咽癌细胞株多药耐药的研究
MDR-reversing effect of (-)epigallocatechin-3-gallate on human nasopharyngeal carcinoma cell lines by decreasing expression of P-glycoprotein
目的 绿茶中的儿茶素单体表没食子儿茶素没食子酸酯( epigallocatechin gallate, EGCG)人耐药鼻咽癌细胞株多药耐药性的逆转作用及相关机制。方法 采用MTT法确定EGCG的非细胞毒性剂量,以及对HNE1/DDP细胞药物敏感性的影响。同时采用荧光定量PCR(QRT-PCR)及Western blot方法分别检测其产物P糖蛋白的表达。结果 EGCG在低于154.6umol/L时对HNE1/DDP细胞的生长抑制率小于10%。60umol/L、80 umol/L及120umol/L的EGCG均可增加HNE1/DDP细胞对顺铂的敏感性,使顺铂对HNE1/DDP 细胞的IC50由原来的4.22ug/mL降低至3.82ug/mL、2.56ug/mL及1.78ug/mL,其逆转倍数分别为1.10、1.65和2.37倍。Western blot结果显示不同浓度的EGCG均可抑制HNE1/DDP细胞P糖蛋白的表达。结论 EGCG可部分逆转人耐药鼻咽癌细胞株HNE1/DDP对顺铂的耐药性,其逆转机制与增加细胞内化疗药物浓度,抑制P糖蛋白的表达有关。
Objective To evaluate the value of (-)epigallocatechin-3-gallate (EGCG) on human nasopharyngeal carcinoma HNE1/DDP cell lines and its possible mechanisms. Methods Cytotoxicity of EGCG and its sensibility to Cis-platinum(DDP) in HNE1/DDP cell line, were evaluated by Methyl thiazolyl tetrazolium (MTT) assay . Before and after EGCG was given, the expressions of P- glycoprotein were detected by Quantitative real-time polymerase chain reaction (QRT-PCR) and Western blot respectively. Results The 10% inhibition concentration (IC10) of EGCG to HNE1/DDP cell line was 154.6umol/L, so 60umol/L,80umol/L and 120umol/L of EGCG were used to detect the reversal effect of multidrug resistance. IC50 for DDP was 4.22ug/mL .EGCG could increase the drug susceptibility of HNE1/DDP cell to DDP, and at the concentrations of those above .The IC50 for DDP decreased to3.82ug/mL, 2.56ug/mL and 1.78ug/mL, and the modulating factors were 1.10,1.65 and 2.37 fold respectively. The expressions of P-glycoprotein in HNE1/DDP cells incubated with EGCG were deceased when detected by QRT-PCR and Western blot. Conclusion EGCG could partially reverse the multidrug resistance of the HNE1/DDP cells in vitro. Moreover, the reversal effect may be due to the increase of intracellular accumulation of chemotherapeutic drugs and the decrease of the expression of P-glycoprotein .
