Experimental study of HZT extracts on blood glucose and blood lipid of rats with type 2 diabetes mellitus

目的 研究降糖孜亚比提片(Hypoglycemic Ziyabiti Tablet,HZT) 对2型糖尿病(T2DM)大鼠血糖和血脂的影响及机制。方法 采用高脂乳剂给Wistar大鼠灌胃10天后,用毛细管法判定胰岛素抵抗性;再结合两次给药法腹腔注射四氧嘧啶(alloxan,ALX),建立T2DM大鼠模型。动物分为正常对照组、模型组、HZT给药组(440、220 mg•kg-1•d-1)及阳性对照消渴丸组(420 mg•kg-1•d-1)。结果 正常对照组与模型组大鼠胰岛素敏感性指数(KITT值)分别为5.8±0.5和 2.8±0.9,模型组明显低于正常对照组(p﹤0.01)。与模型组比较,HZT高、低剂量组均可降低T2DM大鼠的空腹血糖(FBG)和血清胆固醇(TC)水平,能提高高密度脂蛋白(HDL-C)水平,差异具有统计学意义(P﹤0.05);HZT高剂量组可提高空腹血清胰岛素(FINS)水平,差异具有统计学意义(P﹤0.05);HZT高、低剂量组均可使胰岛素分泌指数(IS)增大(P﹤0.05)。结论 HZT能降低T2DM大鼠的FBG和TC水平,其降血糖机制可能是通过促进胰岛素分泌、增加细胞对葡萄糖的利用而起作用的。

im To study the effects of Hypoglycemic Ziyabiti Tablet (HZT) extracts on blood glucose 、blood lipid and metabolism in type 2 diabetic rats. Methods After Wistar rats were administrated with high fat emulsion for 10 days, we assessed the insulin resistance by using blood capillary method. They were injected alloxan (ALX) to abdominal cavity in continuous two days , and then type 2 diabetic rat model was established . The rats were divided into normal control group,model group,HZT treated group(440、220 mg•kg-1•d-1) and positive control Xiaoke Wan (XKW) treated group(420 mg•kg-1•d-1). Results The KITT (exponent of insulin sensitivity) of Wistar rats was 5.8±0.5 in normal control group and 2.8±0.9 in model group (P﹤0.01). Compared with model group, the FBG and TC level in the high-dose and low-dose HZT treated group were decreased and the HDL-C level was increased (P﹤0.05); the FINS level in the high-dose HZT treated group was increased (P﹤0.05) ; the number of IS in the high-dose and low-dose HZT were enlarged (P﹤0.05) . Conclusion HZT could decrease the T2DM rats’FBG and TC level , which hinted the hypoglycemic mechanism was possibly to promote insulin secretion and increase the utilization of glucose .
