目的:探讨FoxE1基因启动子甲基化和结直肠癌发生的关系。方法:对19例I期结直肠癌组织及其配对的正常结直肠粘膜上皮组织,采用实时定量甲基化PCR进行FOXE1基因的甲基化检测,比较结直肠癌和正常结直肠粘膜上皮的FOXE1甲基化水平差异,探讨FOXE1基因甲基化和患者临床病理特征的关系。结果:FOXE1甲基化阳性率为63.2%(12/19),配对的正常结直肠粘膜上皮组织均为非甲基化(0/19),结直肠癌的FOXE1甲基化水平显著高于正常粘膜上皮组织(p=0.029)。患者性别、年龄、肿瘤部位、组织学类型、分化程度、浸润深度、脉管侵犯均和FOXE1基因启动子甲基化状态没有显著性关系。结论:FOXE1启动子甲基化具有癌特异性,在结直肠癌中的甲基化水平显著高于正常结直肠粘膜上皮组织。

Objective:To explore the relationship between methylation of FOXE1 gene promotor and tumorigenesis of colorectal cancer.Methods: This study assessed the methylation of FOXE1 on 19 specimen of stage I colorectal cancer and 19 paired specimen of normal colorectal mucosa using realtime quantitative methylation-specific PCR, compared the difference on methylation of FoxE1 between normal colorectal mucosa and colorectal cancer, and explored the relationship between methylation of FOXE1 gene promotor and the clinicopathological features of colorectal cancer.Results: The positive rate of FOXE1 methylation is 63.2%(12/19)in stage I colorectal cancer, while none of the samples from normal colorectal mucosa was positive. The level of FOXE1 methylation of stage I colorectal cancer is significantly higher than that of normal colorectal mucosa (p=0.029). There are no significant correlation between the status of the FOXE1 gene promotor and patient gender, age, location of tumor, histological type, degree of differentiation, invasive depth or vascular invasion.Conclusion: The level of FOXE1 methylation of stage I colorectal cancer is significantly higher than that of normal colorectal mucosa. FoxE1 gene promotor methylation is cance specific in colorectal cancer.
