NA甲基转移酶抑制剂在急性髓系白血病中的应用与进展
Progress of DNA Methyltransferases Inhibitors Treatment In Acute Myeloid Leukemia
急性髓系白血病作为一种恶性血液系统疾病,极大的危害了人类健康,它是由多种因素导致的疾病,其发病机制之一为相关抑癌基因的DNA甲基化异常。DNA甲基转移酶是基因组甲基化的关键酶,在表观遗传调控基因表达中起着重要作用。阿扎胞苷和地西他滨作为临床治疗中使用的DNA甲基转移酶抑制剂,通过对DNA去甲基化作用治疗急性髓系白血病,本文总结了临床应用中阿扎胞苷和地西他滨在治疗急性髓系白血病时的作用及其临床应用,为个性化治疗和指导临床合理用药提供理论基础。此外,总结国内外对DNA甲基转移酶抑制剂在白血病作用机制研究中的进展,为其表观遗传机制的深入研究和靶向治疗的应用提供了思路和方向。
cute myeloid leukemia(AML) is a malignant blood diseases and it has great harm to human health. DNA methylation mainly refers to the abnormal genomic CpG island methylation, which occurs abnormal gene expression. DNA methyl transferase is a key enzyme, which plays an important role in epigenetic regulation of gene expression and inhibition. Through the possible relationship between DNA methylation molecular abnormalities and AML, it occurrs analysis describes the possible pathogenesis of AML. In recent years, the studys found that some tumor suppressor genes and tumor-related gene hypermethylation are closely related to development of AML. For example RIL, ID4, SFRP family and so on. The mechanism using 5-azactidine and Decitabine treatment of AML is unknown and the use-method is different. Further investigation will afford theoretical principle for individualized treatment. AML is a disease caused by various factors, whose direction of the molecular target is hoping for further treatment breakthrough , and may be considered in combination use of medicine.
