【目的】研究调节性B细胞(regulatory B cells, Bregs)对矽肺纤维化Th免疫应答的影响。【方法】健康成年雌性C57BL/6小鼠,按体重排序后随机分为对照组、矽肺组和Bregs消除组。模型组和Bregs消除组小鼠行非暴露式气管内灌注二氧化硅悬液。Bregs消除组小鼠腹腔注射anti-CD22单克隆抗体消以除小鼠体内Bregs。分别于灌注后7、28及56天,处死小鼠,收集肺组织及肺泡灌洗液上清。测定肺组织羟脯氨酸含量。采用酶联免疫吸附法检测肺泡灌洗液上清中的细胞因子。【结果】二氧化硅灌注后56天,Bregs消除组小鼠肺组织羟脯氨酸含量显著低于矽肺组小鼠(p<0.05)。二氧化硅灌注后28天,Bregs消除组小鼠IL-2的分泌水平显著高于对照组小鼠及矽肺组小鼠(p<0.05)。二氧化硅灌注后56天,Bregs消除组小鼠IL-13的分泌水平显著低于矽肺组小鼠(p<0.05)。【结论】Bregs可通过影响Th1/Th2免疫平衡促进矽肺小鼠胶原沉积。

Objective] To observe the effect of regulatory B cells (Bregs) on Th immune responses in silica-induced lung fibrosis in mice. [Methods] Healthy adult female C57BL/6 mice were divided into 3 groups: control group, silicosis group and Bregs depletion group. To setup the silicosis model, mice in silicosis group and Bregs depletion group received 3mg/50μl silica suspension intratracheally. To deplete Bregs, mice in Bregs depletion group were injected intraperitoneally with 300 μg anti-CD22 Abone day before the silica exposure. Mice were sacrificed at 7, 28 or 56 days after silica exposure. Bronchoalveolar lavage fluid and lung tissues were obtained. The hydroxyproline content in mice lungs and the secretions of cytokines IL-2 and IL-13 were examined. [Results] The hydroxyproline content of lung in Bregs depletion mice decreased obviously at day 56 after silica exposure compared with that in silicosis group (p<0.05). The secretion of IL-2 in Bregs depletion mice increased significantly at day 28 after silica exposure compared with that in control group and silicosis group (p<0.05). The secretion of IL-13 in Bregs depletion mice decreased significantly at day 56 after silica exposure compared with that in silicosis group (p<0.05). [Conclusion] Bregs promoted the collagen deposition in mice lung through affecting Th1/Th2 immune balance.
