二氧化锰纳米酶作为载体递送反义寡核苷酸的研究
Manganese dioxide nanozymes as carriers for antisense oligonucleotides delivery
本文制备了氨基化糖原和牛血清白蛋白修饰的二氧化锰纳米酶,并探究了其作为反义寡核苷酸(Smyd3-ASO)递送载体的可行性和有效性。首先制备了牛血清白蛋白修饰的二氧化锰和氨基化糖原,并将二者交联合成具有正电性的纳米酶载体(NGBM)。NGBM通过静电作用装载Smyd3-ASO。实验结果表明NGBM具有谷胱甘肽氧化酶(GSH-OXD)和过氧化物酶(POD)活性,且具有较高的Smyd3-ASO装载量。流式分析结果表明NGBM可通过巨胞饮途径显著提高Smyd3-ASO的细胞摄取;细胞毒性实验结果表明NGBM/Smyd3-ASO可有效抑制肿瘤细胞生长。
In this study, amino glycogen and bovine serum albumin functionalized manganese dioxide nanozyme was prepared and itsfeasibility and effectiveness as vector for antisense oligonucleotides (Smyd3-ASO) deliverywas investigated. BSA functionalized manganese dioxide andaminated glycogen were synthesized, respectively, and then crosslinked to prepare the positive nanozyme carrier (NGBM). Smyd3-ASO was loaded on NGBMthrough electrostatic interactions. The results showed that NGBMexhibited glutathione oxidase (GSH-OXD) and peroxidase (POD) activities, and had a high Smyd3-ASOloading capacity. Flow cytometry analysis showed that NGBM could significantly improve the cellular uptake of Smyd3-ASO mainlyby macropinocytosis. Cytotoxicity assay showed that NGBM/Smyd3-ASO could effectively inhibit tumor cells growth.
