目的:探索miR-519c-5p在食管癌组织和正常组织中的表达,并分析miR-519c-5p对食管癌细胞株生物学功能的影响。方法:运用MTT和克隆形成实验检测miR-519c-5p对细胞增殖的影响,流式细胞术检测miR-519c-5p对细胞周期的影响,实时定量PCR检测食管癌组织中miR-519c-5p表达水平。结果:miR-519c-5p在食管癌组织中表达为4.547122±5.313108,癌旁组织1,瞬时转染miR-519c-5p相对于对照组增殖能力增强,而且克隆数也增多。结论:miR-519c-5p在食管癌组织中的表达异常,癌组织明显高于癌旁组织,miR-5519c-5p对食管癌细胞株增殖明显的促进作用,其在肿瘤发生过程中很可能促进肿瘤发生。

In Objectives :This research was undertaken to explore the expression pattern of miR-519c-5p in esophageal cancer and noncancerous paired specimens, the effect of miR-519c-5p on the EC-109 esophageal cancer cells. Methods :We conducted RT-PCR to quantify the expression of miR-519c-5p in esophageal cancer and noncancerous paired specimens. Functional assays were respectively employed by proliferation, cell cycle assays. Results: The relative expression of miR-519c-5p in esophageal cancer samples was 4.547122±5.313108 which was significantly higher than 1 of adjacent normal ones (P <0.05). MTT assay revealed that miR-519c-5p mimics significantly promotes proliferation of tumor cell at 48 h after transfection at the concentration of 50 nmol/l; Esophageal cancer cells in G0/G1 stage were significantly decreased, cells in S stage were greatly increased after the treatment with 50 nmol/L miR-519c-5p mimics for 48 h in comparsion with control, P<0.05; For cells in G2/M stage, no significant difference was noted, P>0.05. Conclusions:miR-519c-5p is aberrantly expressed in esophageal cancer, esophageal tumor specimens show increased miR-519c-5p expression compared to their normal counterparts. As miR-19c-5p promotes cell proliferation and esophageal cancer cells in G0/G1 stage, miR-19c-5p might act as oncogene in esophageal cancer.
