温度控制在蛋白质结晶中的应用

蛋白质分子的三维结构信息主要通过X射线单晶衍射技术获得,生长蛋白质晶体是X射线衍射技术解析蛋白质分子结构的主要瓶颈问题之一。因此,获得高质量的蛋白质晶体对于结构解析十分重要。影响蛋白质结晶的因素很多,这些因素可以归结为两个方面的影响:(1)对可否结晶的影响,(2)对晶体质量的影响。温度控制是蛋白质结晶的重要影响因素,但在实践中并未受到广泛重视。本文总结了温度对可否结晶和晶体质量的影响,分析认为温度因素是提高结晶筛选成功率和晶体质量工作中应当考虑的重要方面。

Obtaining high quality protein crystal is one of the bottlenecks of structural determination by X-ray crystallography. Factors affecting the crystallization process of protein can be categorized into two groups: (1) those that affect the crystallizability, and (2) those that affect the crystal quality. Temperature is an important factor for protein crystallization, but it does not attract much attention in practical crystallization. This paper reviewed the past investigations and recent progresses on the effect of temperature on protein crystallization, and the applications of utilizing temperature control were also summarized and discussed. The temperature control was then proposed as an essential factor to be considered in terms of enhancing crystallizability and crystal quality.
