叶柄粘球菌STXZ77菌株抗肿瘤活性物质的初步分离
Preliminary separate the antitumor activity compouds of Myxococcus stipitatus STXZ77
通过肿瘤细胞毒性试验研究了STXZ77菌株的抗肿瘤生物活性,其发酵上清液对B16、HCT-16细胞具有高效的抑制效果,具有剂量依耐性。对发酵上清液利用温度梯度处理后进行肿瘤细胞毒性实验,结果表明发酵液上清中的主要抗肿瘤活性物质具有热不稳定性。利用硫酸铵分级沉淀提取发酵上清液中的蛋白,检测各饱和度区间沉淀蛋白的肿瘤细胞毒性,结果显示,40% ~ 60%饱和度区间的细胞毒性最强,80% ~ 100%饱和度区间的细胞毒性最弱。将发酵液上清经80%饱和度硫酸铵沉淀,透析除盐,得到抗肿瘤蛋白粗提物,再经阴离子交换色谱对抗肿瘤蛋白粗提物初步分离,为抗肿瘤活性蛋白组分的进一步分离奠定了重要基础。
In this study, the antitumor activity of strain STXZ77 was studied by toxicity test. The supernatant of the fermentation broth had a highly effective toxicity effect on B16 and HCT-16 cells in dose-manner. The fermentation supernatant lost antitumor activity in more than 60℃ water-bath heating for half an hour, indicated that the main active substance in the fermentation supernatant with thermal instability. the antitumor activity of precipitation from the fermentation supernatant protein by solid ammonium sulfate fractionation was tested. The results show interval of 40% ~ 60% saturation has the strongest cytotoxicity, while interval of 80% ~ 100% weakest. The protein crude extract was obtained by 80% saturation ammonium sulfate precipitation, and then the antitumor active compound was separated from the crude extract by anion-exchange chromatography, which laid an important foundation for the search of new antitumor drugs.
