褶皱假丝酵母脂肪酶催化合成丁酸香叶酯的优化
Optimization of geranyl butyrate synthesis catalyzed by Candida rugosa lipase
本文研究了褶皱假丝酵母脂肪酶(CRL)在有机相中催化合成丁酸香叶酯的反应,通过单因素实验考察了反应溶剂、酶加量、酸醇底物摩尔比和分子筛添加量对酯化率的影响,通过红外光谱以及核磁对产物进行定性检验,并利用GC测定酯化率,得出了最适反应条件是:反应时间为3.5 h,最适溶剂正庚烷,加酶量9 mg/mL,底物酸醇摩尔1:1.5,分子筛添加量10 mg/mL,在此条件下游离酶催化丁酸香叶酯的酯化率达到91%。
In this work, the synthesis of geranyl butyrate catalyzed by Candida rugosa lipase (CRL) was studied. The influence of the solvent, the enzyme dosage, the molar ratio of butyric acid to geraniol and the amount of molecular sieve on the esterification rate was investigated. The products were identified qualitatively by IR and NMR and quantitatively by GC. It was concluded that the highest esterification rate of 91% was obtained under optimized conditions: 9 mg/mL CRL, 10 mg/mL molecular sieves, 1:1.5 molar ratio of butyric acid to geraniol for 3.5 h in n-heptane.
