小麦与叶锈菌互作过程中H2O2的时空分布特点及其功能分析

H2O2是植物体内的重要信号分子。本实验室前期工作已经证明在叶锈菌侵染小麦诱发的细胞程序性死亡过程中有H2O2爆发。为进一步探讨小麦与叶锈菌互作过程中H2O2产生的时空特点,本试验利用DAB、CeCl3特异性标记H2O2,分别借助光学显微镜及透射电子显微镜来观察叶锈菌侵染过程中H2O2的时空分布特点,以期对H2O2产生及功能进行分析探讨。结果表明,在侵染早期不亲和组合在叶锈菌的侵染结构下泡囊、吸器母细胞和吸器上有H2O2的分布,侵染点被侵染细胞和周围邻近细胞的细胞壁、细胞膜上同样分布大量H2O2 。而亲和组合只在附着胞形成时期的寄主细胞壁上有少量H2O2分布。我们初步推测小麦与叶锈菌互作过程中产生的H2O2主要来源于寄主细胞的细胞膜和细胞壁;在互作早期产生的H2O2可能主要发挥信号作用诱发寄主细胞产生一系列防御反应,而后期产生的H2O2直接参与对菌的杀戮作用和对寄主细胞发生HR的促进作用,同时作为信号物质诱发邻近HR的周围细胞产生防御反应。
H2O2 is an important signaling molecule in plants. Previous work in our lab have been proved through the experiment of pharmacology that H2O2 outbreak is associated with programmed cell death of host cells infected by leaf rust. To further explore the spatial and temporal distribution and the source of H2O2 during the interaction between Triticum aestivum and Puccinia triticina, we used DAB, CeCl3 to mark H2O2 and observed it through optical microscopy and transmission electron microscopy. The results showed that H2O2 mainly distributed on sub-stomatal vesicles , haustorial mother cell and haustorium, within the host cell which infected by leaf rust and on the health mesophyll cell plasma membrane arounding the infected site in the incompatible combination at early stage. But in the compatible combination, H2O2 mainly concentrated on the guard cells of infection point. So,we gusee that H2O2 generated during the interaction sourced mainly from the host cell membrane and wall. At the earlier stage of interaction, H2O2 might play a major role in inducing the host cells to produce a series of defensive response. At the later stage of interaction, H2O2 directly involved in the killing effect on leaf rust and the promoting effect on host cells to produce HR, while as a signal could induce neighboring HR cells to produce defensive reactions.
