厌氧氨氧化菌Ca. Brocadia胞外聚合物的荧光特性及来源探究

本研究旨在探究厌氧氨氧化菌Ca. Brocaida胞外聚合物自发荧光的来源与特性,采用荧光原位杂交、三维荧光光谱、高效液相、标准加入法等技术手段证明其352/464 nm和442/521 nm两种荧光的物质来源分别为细胞代谢物辅酶NADH和核黄素的胞外分泌,并发现Ca. Brocadia型厌氧氨氧化污泥中荧光物质的含量高于硝化污泥以及Ca. Jettenia型厌氧氨氧化污泥。这对于深入探究Ca. Brocadia的代谢机制以及污泥菌群结构的表征具有重要意义。
In this paper, autofluorescence characteristics and source of extracellular polymeric substances released by anammox bacteria Ca. Brocadia was investigated. Coenzyme NADH and riboflavin were verified as the sources of autofluorescence (352/464 nm and 442/521 nm) via fluorescent in situ hybridization, excitation-emission matrix fluorescent spectroscopy, HPLC and standard addition method. And it was found that fluorescent substances contents were higher in Ca. Brocadia than in nitrifying sludge and Ca. Jettenia. It was of great importance for exploring Ca. Brocadia metabolic mechanism and characterizing sludge community structure.
