猕猴桃果实成熟衰老进程中乙烯受体基因家族成员的表达研究

利用猕猴桃EST数据库克隆到5个乙烯受体基因家族成员(AdERS1a,AdERS1b,AdETR1,AdETR2和AdETR3)。在果实生长发育进程中,AdERS1a和AdETR3表达呈下降趋势,而AdERS1b和AdETR2则表现为峰型变化,在花后97 d的果实中表达最强。实时定量PCR结果显示,在猕猴桃果实成熟衰老进程中,AdERS1a,AdETR2和AdETR3表达水平随乙烯积累呈增加趋势,同时受外源乙烯处理显著诱导,AdETR1则受内源和外源乙烯下调,而AdERS1b则对内源和外源乙烯均没有响应。1-MCP处理效果均显著延缓了乙烯受体基因家族对乙烯的响应。研究认为猕猴桃果实乙烯受体基因家族在果实成熟衰老进程中具有不同表达,它们可能通过协同作用方式,在感受乙烯的同时将乙烯信号传递给下游元件,进而激活整个乙烯信号转导途径。
Five ethylene receptor gene family members were cloned from kiwifruit EST database. Real-time and endpoint PCR were used to analyze gene expression patterns during development and post-harvest ripening and senescence. During fruit development, levels of transcripts of AdERS1a and AdETR3 decreased, whereas those of AdERS1b and AdETR2 peaked at 97 days after full bloom (DAFB). In ripening kiwifruit, there was a diverse response of the ethylene receptor family to internal and external ethylene. AdERS1a, AdETR2 and AdETR3 expression increased at the climacteric stage and transcripts were induced by external ethylene treatment, while AdERS1b showed no response to ethylene. AdETR1 was negative regulated by internal and external ethylene in ripening fruit, a result not found previously in this receptor family in ripening fruit. 1-MCP treatment prevented the ethylene response of all ethylene receptor genes, but transient down-regulation was only found in AdETR2. The differential ethylene response of the receptors suggests different sensitivities and ethylene binding amongst the family members in co-operating the first stages of the ethylene signal transduction pathway.
