大白菜EMS诱变结合小孢子培养获得突变体

本试验以5个不同基因型大白菜为试验材料,研究了不同浓度EMS浸泡花蕾不同时间对小孢子发育以及产胚率和成苗率的影响。结果表明,EMS浓度高于0.1%的处理均会导致小孢子致死。EMS浓度为0.03-0.1%、处理时间5-10min时,小孢子的产胚率和成苗率较高,比较适合花蕾诱变后进行小孢子培养。通过对小孢子植株生殖生长时期性状调查,筛选出了在花色、叶形、对霜霉病抗性增强的突变株。本研究证明利用EMS诱变大白菜花蕾快速获得再生植株是可行的,从而为快速创建大白菜新种质奠定了基础。
In this study, five different genotypes of Chinese cabbage were used for researching influences on microspore development, production rate of embryo and seedling rate by soaking buds using ethyl methane sulphonate (EMS) with different doses and different duration. The results indicated that microspores wound be lethal when the dose was higher than 0.1%, the production rate of embryo and seedling were high when buds were treated with 0.03 ~ 0.1% of EMS for 5 ~ 10 min and it is suitable for microspore culture after EMS mutagenic buds. The reproductive growth of homozygous plants were investigated, and mutants of flower color, leaf shape, and the resistant mutants of downy mildew were obtained. The results suggested that it is feasible to obtain regeneration plantlets by EMS induced mutation in Chinese cabbage, and provided a basis for creating the new germplasm of Chinese cabbage rapidly.
