3-乙酰乌头碱油浴模拟铁棒锤砂炒炮制工艺参数研究
Study the processing technological parameter of Aconitum pendulum Busch by the methods of using oil bath to imitate 3-acetylaconitine fry processing
目的:筛选3-乙酰乌头碱在砂炒过程中含量发生转化温度和时间的参数范围。方法:采用油浴加热方式模拟砂炒炮制过程,设置120℃、140℃、160℃、180℃,4个温度梯度,每个温度梯度反应1、10、20、30、40 min;采用HPLC法对不同温度、时间反应得到的3-乙酰乌头碱炮制样品进行分析,色谱柱: COSMOSIL PAK 5C18 -MS-Ⅱ(4.6 mm×250 mm,5 μm),流动相:甲醇-0.03mol/L碳酸氢铵溶液(浓氨水调pH9.5)(70:30),等度洗脱;流速:1mL/min;检测波长:230nm;外标一点法计算含量。结果: 随着温度的升高及时间的延长,3-乙酰乌头碱含量发生降低,炮制前3-乙酰乌头碱含量为160μg/mL,在160℃加热30 min后,含量降为65.42μg/mL,加热40 min后,含量降为4.53μg/mL,180℃加热10min后,含量降为27.67μg/mL,20 min时,含量降为0μg/mL。结论: 3-乙酰乌头碱在模拟砂炒炮制过程中发生转化的温度和反应时间参数为160℃~180℃,炮制时间为20~40 min,这一参数能够为下一步铁棒锤砂炒工艺提供一定的参考依据。
Objective: To screen processing translative temperature and time parameters of 3-acetylaconitine content. Methods: Using oil bath to imitate fry processing,setting up four temperature gradient: 120℃, 140℃, 160℃, 180℃, every gradient process 1, 10, 20, 30, 40 min, different temperature and time samples be analyzed by High Performance Liquid Chromatography (HPLC) on a COSMOSIL PAK 5C18 -MS-Ⅱ(4.6 mm×250 mm,5 μm) by isocratic elution with methyl alcohol-0.03mol/L ammonium bicarbonate (adjusted pH9.5) (70:30) . The flow rate was 1mL/min and the detection wavelength was setted at 230nm, using external standard one point method to calculate content. Results: With the increase of temperature and time, the content of 3-acetylaconitine decreace to 65.42 μg/mL when 160℃heat 30 min, when 180℃heat 10 min, decreacing to 27.67 μg/mL, at 20 min, decreacing to 0 μg/mL. Conclusion: the processing technology parameters of temperature is 160℃~180℃, as to the time is 20~30 min, this parameter can provide certain reference basis for the furtherstudy.
