Survivin mRNA在肾透明细胞癌患者组织和外周血中表达的研究

Objective: To study the expression and significance of Survivin mRNA in tumoral tissues and blood cells of patients with renal clear cell carcinoma（RCCC）.Methods: Survivin mRNA in tumoral tissues of 46 patients with RCCC, 15 normal renal tissues, blood cells of 46 patients with RCCC, and 15 healthy volunteers were detected. Survivin and β-actin were amplified by using RT-PCR and RT-PCR-ELISA. Results: 21 of 46 (45.7%) showed positive Survivin mRNA signals in blood in patients with RCCC. Only 2 of 15 (13.3%) patients in control group showed positive Survivin mRNA signals in blood. The expression of Survivin mRNA difference between experimental group and control group was significant (P <0.05). Detection of Survivin mRNA expression in tissue and blood in 46 patients with RCCC at the same time, The positive rates of Survivin mRNA in tumor tissue and blood are 63.0% and 45.7% ,respectively (P<0.01).Conclusions: Carcinoma tissues of the patients with RCCC showed significantly positive expression of Survivin mRNA. High sensitivity and the specificity of expression of blood Survivin mRNA to provide a simple, microinvasive diagnostic test to identify patients with RCCC .The RT-PCR-ELISA is the effective method to detection of Survivin mRNA in the blood of patients with RCCC.
