高效液相色谱法测定蒙古栎橡碗提取物中鞣花酸的含量

目的:建立高效液相色谱法(HPLC)测定蒙古栎橡碗中鞣花酸的含量。方法:采用回流提取法,以乙腈-0.2%磷酸水溶液为流动相,等度洗脱,流速1.0 mL/min,检测波长254 nm,柱温30 ℃,进样量10 μL。结果:鞣花酸含量测定的线性浓度范围在5~100 μg/mL(r=0.9997),平均回收率为100.5%,RSD为2.4%(n=3)。结论:该方法简单,准确,重现性好,可用于蒙古栎橡碗提取物中鞣花酸含量的分离鉴定。
Objective: In order to provide a scientific basis for the determination of ellagic acid in Mongolian oak cups, high-performance liquid chromatography (HPLC) was used in this study. Method: The oak cups were extracted by methanol through heating reflux and the content of ellagic acid was determined by HPLC. The mobile phase was acetonitrile and 0.2% phosphoric acid solution at a flow rate of 1.0 mL/min. The detection wavelength was set at 254 nm, the column temperature was 30 ℃ and injection volume was 10 μL. Result: A good linearity of ellagic acid was achieved in the range of 5~100 μg/mL (r=0.9997). The average recovery was 100.5% and RSD was 2.4% (n=3). Conclusion: This method is simple, accurate and reproductive that can be used as quality control method for ellagic acid of oak cups extractions.
