太湖蓝藻微囊藻毒素的薄层层析研究

微囊藻毒素是太湖蓝藻产生的有毒物质,会对肝脏产生毒害作用。本文以介电常数为依据筛选层析剂组成,采用单因素扫描和全因子设计响应面优化方法获得薄层层析微囊藻毒素MC-LR、MC-RR的展开剂体系溶剂比例为三氯甲烷:甲醇:水=65:25:2。MC-LR、MC-RR的展开斑点通过碘蒸气显色的目测检出最低浓度为1mg•L-1。薄层色谱扫描法捡出的MC-LR、MC-RR最低浓度为0.001mg•L-1。MC-LR、MC-RR的色谱扫描峰面积与其浓度在 0.05~10 mg•L-1 浓度范围内呈线性关系,检测准确度达95%以上。

Microcystins are a series of hepatotoxins produced by Cyanobacteria. According to various dielectric constants of developing solvents, thin-layer chromatography method was developed for detecting the microcystin MC-LR, MC-RR. The single factor test and response surface analysis were introduced for obtaining the optimum ratio of developing solvents is trichloromethane : methanol : water = 65:25:2. The lowest concentration of MC-LR, MC-RR visually observed using the iodonium development is 1mg•L-1. The lowest concentration of MC-LR, MC-RR distinguished by TLC-scanning determination is 0.001 mg•L-1. The peak values of MC-LR、MC-RR are in a linear relationship to the concentrations of them in the range of 0.05~10 mg•L-1. The accuracy of the detection is above 95%.
