不同氟烷基因型猪肉质与能量代谢的关系
Effect of halothane genotype on energy metabolism in longissimus dorsi muscle
本实验选用体重为65 kg左右氟烷基因显性纯合子(HalNN)、杂合子(HalNn),和隐性纯合子(Halnn)的猪作为实验动物,比较不同氟烷基因型猪背最长肌肉质指标,以及血液和肌肉代谢相关指标的差异,以探讨氟烷基因型对肌肉能量代谢的影响,并分析这些指标与肉质之间的关系。结果表明肉质指标HalNN型猪总体优于Halnn型猪,Halnn型猪呈轻度PSE肉症状, HalNn型猪的肉质介于两者之间。HalNN、HalNn型猪背最长肌的糖原含量显著低于Halnn型猪(P < 0.05),HalNN型与HalNn型之间没有差异;HalNn、Halnn型猪背最长肌的乳酸含量显著低于HalNN型猪(P < 0.05),HalNn型与Halnn型之间没有差异;HalNN、HalNn型猪背最长肌CK活性极显著高于Halnn型猪(P < 0.01),HalNN和HalNn型之间没有差异;HalNN、HalNn型猪背最长肌AMPK活性显著低于Halnn型猪(P < 0.05),HalNN和HalNn型之间没有差异。HalNN型猪背最长肌GLUT4 mRNA的表达显著高于HalNn、Halnn型猪(P < 0.05),HalNn和Halnn型之间没有差异;HalNN型猪背最长肌PGC-1α mRNA的表达显著高于Halnn型猪(P < 0.05),HalNn介于两者之间。以上结果提示,在屠宰初期,Halnn型猪由于ATP消耗过快,启动代谢相关的酶,糖原无氧酵解加速,生成大量乳酸,导致最终较差的肉质。
In this study, pigs at 65 kg of body weight differing in halothane genotype (HalNN, homozygous wild type; HalNn, heterozygous; Halnn, homozygous mutant) were employed to investigate the influence of halothane genotype on muscle energy metabolism, in association with meat quality. Results indicated that HalNN pigs exhibited the best pork quality, Halnn showed a slight PSE syndrome, and HalNn were the intermediate. Halnn demonstrated significantly higher glycogen and LD contents in longissimus dorsi compared with HalNN and HalNn (both P < 0.05), while no difference was observed between HalNN and HalNn. Besides, there was significantly lower CK, higher AMPK activities in longissimus dorsi of Halnn than those in HalNN and HalNn (all P < 0.05). Real-time PCR results showed that GLUT4 and PGC-1α transcripts were significantly higher in HalNN compared with Halnn, and no significent difference between HalNn and Halnn was observed. These results indicate that longissimus dorsi muscles from different halothane genotype exhibit distinct metabolic patterns. Also indicate that HalNN pigs have much higher capacity of energy metabolism both in aerobic metabolism and anaerobic glycolysis which may contribute to the better meat quality.
