益髓通经方对实验性自身免疫性神经炎大鼠坐骨神经传导速度及超微结构的影响
Effect of Yisui Tongjing prescription on the conduction velocity and microstructure of the sciatic nerve in Experimental Autoimmune Neuritis(EAN) rats
目的:观察益髓通经方对格林-巴利综合征(GBS)经典动物模型-实验性自身免疫性神经炎(EAN)大鼠坐骨神经传导速度及超微结构的影响。方法:采用P0180-199多肽免疫Lewis大鼠制备EAN动物模型,随机分为模型组、益髓通经方高、中、低剂量组、免疫球蛋白治疗组5组,另设空白对照组1组。分别给予不同剂量益髓通经方溶液灌胃及免疫球蛋白腹腔注射治疗。在免疫后第10、18、25、39天采用BL-420生物机能实验系统检测各组大鼠坐骨神经运动传导速度;应用电镜观察坐骨神经病理形态学超微结构变化。结果:模型组运动神经传导速度在免疫后第10、18、25天进行性下降,在第39天时略恢复。益髓通经方各治疗组神经传导速度在上述时间点均优于模型组;在第39天时,高剂量组同球蛋白组比较无显著性差异(P>0.05),优于中、低剂量组(P<0.01),但均不及空白组(P<0.01)。电镜观察,模型组在第18天出现有髓神经纤维髓鞘折叠起皱、轴索与髓鞘板层分离等改变,在第25天时出现较明显髓鞘脱失或溶解,在39天时仍有广泛脱髓鞘改变并伴轴索溶解。经治疗,益髓通经方治疗组与球蛋白组的髓鞘脱失情况均有好转。结论:益髓通经方能够提高EAN大鼠坐骨神经运动传导速度,改善有髓神经纤维脱髓鞘改变,促进神经修复和再生。
Objective: To study effect of Yisui Tongjing prescription on the conduction velocity of motor nerve (MNCV) and microstructure of the sciatic nerve in experimental autoimmune neuritis (EAN) rats, the Guillain-Barre Syndrome (GBS) classic animal models. Methods: The Lewis rat models of EAN were made by immunization with P0180-199 and they were randomly divided into 5 groups: the model group, intravenous immune globulin (IVIg) group, high, medium , low dose Yisui Tongjing prescription group. Normal rats were taken as the control group. Each group was treated by different dose of Yisui Tongjing liquid or IVIg injected into the abdominal cavities. MNCV and microstructure of the sciatic nerve were tested with BL-420 biological functional system and electron microscope on the 10th , 18th , 25th , 39th day after immunization.Results: Compared with normal group, the MNCV in model group were significantly decreased on the 10th , 18th , 25th day and little increased on the 39th day. The MNCV in each Yisui Tongjing prescription group was higher than that in model group. On the 39th day, the MNCV showed no significant difference between high dose group and IVIg group, but was higher than that in medium and low dose group(p<0.01), and was lower than that in normal group(p<0.01). Folds and crinkles of myelin sheath and wide gaps between axon and sheath appeared on the 18th day, demyelination and myelinolysis were obvious on the 25th day and most serious on the 39th day in model group. The above changes were significantly ameliorated in high dose group and in IVIg group after treatment.Conclusion: Yisui Tongjing prescription can increase the MNCV of the sciatic nerves in EAN rats, alleviated the demyelination and promote the nerve repair and regeneration.
