目的 观察辛伐他汀是否对动脉粥样硬化(AS)家兔模型核因子-KB(NF-KB)表达有影响并抑制相关炎症因子TNF-α、IL-1和MCP-1的作用。方法 40只雄性日本大耳白家兔随机分成空白对照组、模型组、辛伐他汀组,除空白对照组外,其余各组建立主动脉粥样硬化斑块模型,从实验第1天开始予辛伐他汀早期干预,第10周末麻醉处死后取主动脉行NF-KB免疫组化染色观察,并于实验0,3,6,10周动态检测血清中TNF-α、IL-1及MCP-1含量。结果 模型组粥样斑块中泡沫细胞NF-κBp65亚基表达丰富,辛伐组NF-κBp65亚基较模型组阳性细胞表达减少,对斑块区NF-κB p65亚基的表达有明显的抑制。模型组各时间点血清中TNF-α、IL-1和MCP-1含量显著增高(P<0.01),辛伐给药组在各时间点均有不同程度的降低IL-1、TNF-α和MCP-1水平的作用(P<0.05或P<0.01)。结论 辛伐他汀能减少斑块区NF-κBp65亚基表达,在不同时间点对炎症因子的释放有抑制作用。

Objective To investigate whether simvastatin on atherosclerosis (AS) rabbit model of nuclear factor-KB (NF-KB) expression and inhibit the related impact on inflammatory cytokines TNF-α, IL-1 and MCP-1. Methods 40 male Japanese rabbits were randomly divided into blank control group, model group, simvastatin group, with the exception of the control group, the other in each group to establish aortic atherosclerosis model, from the beginning of the experiment day 1 early intervention to simvastatin, 10 were killed over the weekend after the anesthesia line access aortic NF-KB immunohistochemical staining, and the experimental dynamic 0,3,6,10 weeks in serum TNF-α, IL-1 and MCP-1 content. Model group at different time points in serum TNF-α, IL-1 and MCP-1 concentration was significantly higher (P <0.01), Simvastatin cutting treatment group at each time point in varying degrees to reduce IL-1, TNF-α and MCP-1 level of the role of (P <0.05 or P <0.01). Conclusions Simvastatin reduces NF-κBp65 plaque areas subunit expression, at different time points on the release of inflammatory cytokines inhibit.
