microRNA( miRNA)主要通过引导靶基因mRNA的降解或阻止翻译过程来调节基因的表达,在植物的生长发育或影响对逆境的适应性方面起着重要调控作用。植物胚胎发育是一个极其复杂的过程,其基因的差异表达必然存在miRNA的调控。但是,截止目前为止,有关植物体胚中小RNA的研究尚未见系统报道。本研究第一次对龙眼体胚发生过程进行小RNA文库构建和Solexa测序。研究结果表明:共得到Unique 序列6,553,782条,其中仅13,151条序列与已知的miRNAs相似;sRNA以24 nt的长度作为主要存在方式;龙眼体胚发生过程中miRNA的表达丰度存在巨大差异,从一次到数万次不等,其中大部分为低丰度表达;总共鉴定了367个已知的柑橘miRNA,构成纵多家族,不同家族间成员数量存在巨大差异;另外总共获得23个候选的新miRNA,其中有10个miRNA含单个基因座,另13个miRNA具备多个基因座;69个候选的miRNA预测到了靶基因,总共对应着约70个基因符合靶标特征。

MicroRNAs (miRNAs) play important regulatory roles in various biological and metabolic processes by negatively modulating gene expression at the post-transcriptional level. Plant embryo development is a complex process, whcih must be exist a large number of specific miRNAs expression. However, to date, no systematic studies of small RNA in plant embryo development have been conducted, and there is no report about longan somatic embryo miRNAs.In this study, to investigate the number of miRNAs related to embryogenesis in longan and what features these miRNAs exhibit, Solexa sequecing technology was employed to identify miRNAs from a short RNA library generated from synchronized longan embryogenic cultures at different developmental stages. Solexa sequencing showed that longan has a complex small RNA population and the length of small RNAs varied, 24 nt being the predominant. 12, 554, 858 reads produced 13,151 unique miRNAs representing 367 conserved miRNAs as well as 23 novel poential miRNAs were obtained. The expression patterns of different unique sRNAs during the development of longan somatic embyrogenesis varied drastically, and most of them were at the low levels; Specifically, potential target genes were predicted for some conserved miRNAs based on sequence homology search. This research provides a first large scale cloning and characterization of plants(longan) somatic embryogenesis related miRNAs. The newly identified miRNAs greatly enrich the repertoire of somatic embryo miRNAs and provide insights into miRNA biogenesis and expression in plant somatic embryo development.
