GhIDD57调控棉花黄萎病抗性机理研究
Mechanism Research of Cotton Resistance to Verticillium Wilt regulated by GhIDD57
黄萎病严重危害棉花生产,解析棉花抗黄萎病机制具有重要意义。INDETERMINATE DOMAIN(IDD)家族是植物特异性转录因子家族,课题组前期研究发现GhIDD57具有负调控棉花黄萎病抗性的功能,但是具体的调控机制尚不清楚。本研究利用病毒诱导的基因沉默(VIGS)技术在棉花中沉默GhIDD57,对沉默植株叶片转录组测序,结合生物信息学分析GhIDD57结合的启动子基序,筛选到GhMYB3作为其下游靶基因。利用RT-qPCR在GhIDD57沉默植株中检测到GhMYB3的表达量显著升高,在GhIDD57过量表达的棉花原生质体中检测到GhMYB3的表达量显著下降,酵母单杂交分析结果表明GhIDD57可以直接结合GhMYB3启动子motif并抑制GhMYB3的表达,证明GhMYB3在GhIDD57下游并受其负调控。
Verticillium wilt seriously harms cotton production, so it is of great significance to analyze the mechanism of resistance to verticillium wilt in cotton. The INDETERMINATEDOMAIN (IDD) family is a family of plant-specific transcription factors. Our previous study found that GhIDD57 can negatively regulate cotton verticillium wilt resistance, but the specific regulatory mechanism remains unclear. In this study, virus-induced gene silencing (VIGS) technique was used to silence GhIDD57 in cotton, and the silenced plant leaves were sequenced, and the GhIDD57 binding promoter motif was analyzed with bioinformatics, and GhMYB3 was screened as its downstream target gene. The expression of GhMYB3 was significantly increased in GhIDD57 silenced plants and significantly decreased in cotton protoplasts with overexpression of GhIDD57 by RT-qPCR. The results of yeast single hybridization analysis showed that GhIDD57 can directly bind to GhMYB3 promoter motif and inhibit GhMYB3 expression, indicating that GhMYB3 is downstream and negatively regulated by GhIDD57.
