新一代测序技术(Next Generation Sequencing,NGS)又称为高通量测序技术,是DNA测序史上一次划时代的革命。新一代测序技术都采用了一种新的测序策略--循环芯片测序法(cyclic-array sequencing),可分为焦磷酸测序技术(Roche/454)、Solexa测序技术(Illumina)、polony测序技术(ABI/SOLiD)。由于其高通量、成本低、耗时短而迅速取代传统的Sanger测序成为后基因组时代的重要研究手段,让研究者能以更低廉的价格,更全面、更深入地分析基因组、转录组及蛋白质之间交互作用组。新一代测序技术除了可以快速实现肿瘤基因组测序外,还是数字化全面快速剖析肿瘤转录组、表观遗传及DNA-蛋白质交互作用的有力工具,有望在生命科学、医学科学等领域带来革命性的变革。

he next-generation sequencing (Next Generation Sequencing, NGS), also known as high-throughput sequencing technology, is a landmark of revolution in the history of DNA sequencing. Next-generation sequencing technology have adopted a new sequencing strategy-cyclic-array sequencing, divided into pyrosequencing (Roche/454), Solexa sequencing technology (Illumina) the polony sequencing technology (ABI/ SOLiD), Polonator sequencing technology and HeliScope single-molecule sequencing technology. Because of its high-throughput, low cost and short time-consuming it has replaced traditional Sanger sequencing became the important research tools in post-genomic era. Researchers could do more comprehensive and in-depth analysis with genome, transcriptome and proteome at cheaper prices. Next-generation sequencing has broad application prospects in the field of cancer research, in addition to quickly realize the tumor genome sequencing, but also provide us a fully digital rapid powerful tool for tumor transcriptome, epigenetic and DNA-protein interaction analysis. It was expecting to bring about revolutionary change in the life sciences, medicine science and other fields.
