目的:确定超顺磁性氧化铁纳米颗粒Resovist标记大鼠骨髓间充质干细胞的有效性和安全性。方法:分离培养鉴定大鼠骨髓间充质干细胞(MSCs),用Resovist 50μg/ml共孵育24h,通过普鲁士蓝染色、细胞铁浓度测定、细胞透射电镜检查观察细胞的标记效率,细胞增殖试验和细胞活力检查观察细胞的标记毒性。结果:(1)大鼠MSCs经Resovist标记后,普鲁士蓝染色胞内均可见大量着色颗粒,标记率接近100%,电镜检查显示胞质内含铁致密颗粒囊泡,电镜下细胞微细结构未见氧化铁的破坏和毒性作用,原子吸收分光光度法测定细胞平均铁含量为21.77±3.62 pg。(2)Resovist标记和外加磁场对细胞增殖能力、细胞活力均无明显影响。结论:Resovist能高效、安全标记大鼠骨髓MSCs,在细胞磁靶向方面具有动物实验研究和临床应用潜力。

Objective:To explore the effectiveness and safty of labeling rat bone marrow mesenchymal stem cells(MSCs) with Resovist, a kind of superparamagnetic iron oxides(SPIO).Methods:MSCs isolated from Sprague-Dawley rat were incubated with Resovist nanoparticles at the concentration of 50 μg/ml for 24 hours.The labeling effectiveness of Resovist particles in MSCs was assessed by Prussian BIue staining, electron microscopy and atomic absorption spectrometry.The Cell toxity of Resovist particles was observed by the proliferation assays and cellular viability with the help of a CASY?2 Analyser.The cellular effect of a 0.6T magnetic fields on MSCs were also assessed. Results:The Prussian blue staining of the MSCs showed intracytoplasmic iron inclusions as dense blue-stained vesicles, and the labeling efficiency was approximately 100%. Transmission electron microscopy of labeled cells indicated the presence of anionic magnetic nanoparticles exclusively in polydisperse vesicles in the cytoplasm, and no obvious change in the ultramicrostructure was observed. The average cellular iron content was 21.77 ± 3.62 pg per cell after 24 hours of incubation. The proliferation rate and viability rate showed no differences between unlabeled MSCs and labeled MSCs with and without exposure to magnetic fields. Conclusion:Resovist could effectively and safely label rat bone marrow stem cells, which may be useful in cellular magnetic targeting strategies.
