本文利用单克隆抗体、部分酸水解和NMR等方法研究了两种I-型聚鼠李半乳糖醛酸(RG-I)人参果胶的精细结构。结果表明,这两种RG-I型人参果胶的主链均由[→4)-α-GalA-(1→2)-α-Rha-(1→]二糖重复结构单元组成,具有中性糖和酸性糖侧链。中性糖侧链结构均含阿拉伯聚糖、半乳聚糖和I-/II-型阿拉伯半乳聚糖,通过Rha的C-4位连接到RG-I的主链上。酸性糖侧链通过Ara或Gal连接到RG-I的主链上,但酸性糖侧链的GalA聚合度在两种RG-I型果胶中不同。研究结果对于深入理解RG-I型人参果胶的结构及其果胶药理活性的构效关系具有重要的意义。

In this paper, we have employed monoclonal antibodies in combination with acid hydrolysis and NMR spectroscopy to assess the fine structures of two ginseng rhamnogalactouronan-I (RG-I) domains. Our results indicate that both RG-I species have typical RG-I backbone structures with [→4)-α-GalA-(1→2)-α-Rha-(1→] repeat units, respectively, in addition to neutral and acidic side chains. The neutral side chains in both species are similar with arabinan, galactan, and AG-I/II structures linked to the RG-I backbone via C-4 of Rha residues. However, the lengths of their acidic GalA moieties, linked via side chain Ara and Gal sugars, are different. Our studies provide significant insight into the chemical structure of RG-I type pectins, information that may impact on their function in the development of pectin-based therapeutics.
