参与葡萄糖醛酸代谢的尿苷二磷酸葡萄糖醛酸转移酶(UGT)是Ⅱ相代谢中最重要的酶,催化多种内源和外源性物质的代谢反应,对维持机体稳态平衡具有重要作用。UGTs之间的相互作用形成寡聚体后会在较大程度上改变酶的活性,而引起代谢能力的改变,甚至导致药物中毒。因此,研究UGTs之间的蛋白-蛋白相互作用及作用机制,可以为临床上实现个体化合理给药提供科学依据。本文综述了UGTs同源及异源寡聚体的形成及其机理的研究现状。
UDP-glucuronosyltransferases (UGTs) are the most important drug phase II enzymes, which catalyze a variety of endogenous and exogenous substances metabolic reactions. They play important role in maintaining homeostasis. The formation of UGTs oligomers will have an effect on the activity of the UGTs enzyme in a large extent, which leads to changes in metabolic capacity, and even drug intoxication. Therefore, it is of great significance for personalized medicine to study the protein-protein interactions between UGTs. This review summarizes the advances in studies on the formation of UGTs oligomers and the mechanism.
