目的:为了探索miR-181a是否受TGF-β诱导及其对肝癌转移的作用。 方法:通过real-time PCR检测TGF-β处理后的肝癌细胞中miR-181a的表达情况,进一步探讨了其作用机制;同时还检测了肝癌标本中miR-181a的表达并分析了其临床意义;此外通过transwell实验方法检测了miR-181a对肝癌细胞侵袭能力的影响。结果:Real-time PCR结果显示TGF-β直接诱导miR-181a的表达并依赖于Smad2途径;miR-181a在肝癌组织中的表达高于癌旁和正常肝组织,且与转移密切相关;侵袭实验结果进一步证实miR-181a体外可促进肝癌细胞侵袭。结论:本文揭示了miR-181a的表达受到TGF-β的诱导,且与肝癌转移相关;miR-181a体外可增强肝癌细胞的侵袭能力;以上发现为肝癌的治疗提供了一个新的潜在靶点。

Abstract

Objective: To reveal the response of miR-181a expression to TGF-β presence and the function of miR-181a on hepatocellular carcinoma. Method: The relative expression of miR-181a in HCC cells treated with TGF-β was tested by real-time PCR. Moreover, the expression level of miR-181a in HCC samples and the clinical significance were analyzed. Furthermore, the effect of miR-181a on HCC cell invasion was detected through transwell assays coated with matrigel. Result: TGF-β induces miR-181a expression which was dependent on the Smad2 pathway; miR-181a was upregulated in HCC tissues compared to noncancerous tissues and normal liver tissues, and its expression was correlated with metastasis of HCC. Additionally, miR-181a could promote HCC cell invasion in vitro. Conclusion: In this paper, it was discovered that TGF-β induced miR-181a expression which was related to HCC metastasis and could enhance the HCC cell invasion. Thease findings provide a potential target for HCC therapy.
