UCH-L1在乳腺癌中的表达及其临床意义

目的 研究UCH-L1在乳腺癌中的表达及其与临床病理特征的关系。方法 采用免疫组织化学S-P法检测80例乳腺浸润性导管癌石蜡标本中UCH-L1的表达情况，采用Western blot 检测60例新鲜乳腺浸润性导管癌组织以及癌旁正常乳腺组织标本中UCH-L1的表达情况。结果 免疫组化结果显示在80例乳腺浸润性导管癌中，UCH-L1阳性表达42例（52.5%），定位于细胞浆。Western blot 结果显示UCH-L1在乳腺浸润性导管癌中的表达高于癌旁正常乳腺组织。统计数据均显示UCH-L1蛋白在乳腺癌中的表达，与其组织学分级、TNM分期、淋巴结转移情况相关。 结论 UCH-L1在乳腺癌的进展、转移中发挥重要作用。
Objective To investigate the expression and significance of the Ubiquitin carboxy terminal hydrolase-L1 (UCH-L1) in breast cancer. Method Immunohistochemistry S-P method was used to detect the expression of UCH-L1 in 80 paraffin-embedded breast cancer tissues. And western blot was used to detect the expression of UCH-L1 in 60 fresh resection samples of breast cancer and normal breast tissue beside tumor . Result The immunohistochemistry result indicated that the positive rate of UCH-L1 protein in breast cancer was 52.5%, which located in the cytoplasma. And the western blot result indicated that expression of UCH-L1 protein in breast cancer was significantly higher than those in adjacent tissues(normal breast tissue beside tumor). The expression of UCH-L1 was correlated with clinical histological grade,TNM staging, and lymphatic metastasis in brast cancer. Conclusion The results strongly suggest that UCH-L1 protein play an important role in the development and metastasis of breast carcinoma.
