目的:探究三碘甲腺原氨酸(T3)对高糖毒性引起的RIN-m5F细胞氧化损伤的保护作用。 方法:传代培养RIN-m5F细胞,分为正常对照组、高糖组(44mM葡萄糖)、α-硫辛酸组(200μM),T3作用组(浓度分别为10-5M,10-7M,10-9M),培养72h后,进行以下测定:DCFH-DA探针检测细胞内ROS水平;MTT法测定细胞存活率;丙二醛(MDA)、试剂盒测定细胞内丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性和总抗氧化能力(T-AOC);ELISA试剂盒检测葡萄糖刺激胰岛素的分泌水平(GSIS);RT-PCR检测 MafA、Glut2、GCK mRNA表达量的改变。结果:高糖组与Control组比较,细胞存活率和SOD、T-AOC显著降低,ROS水平和MDA含量显著升高,葡萄糖刺激的胰岛素分泌量减少,MafA、Glut2、GK的mRNA表达量降低, LA的添加可以明显抑制高糖引起的上述变化,而T3除了不能改变高糖诱导细胞GCK基因的表达外均可以抑制其他的变化,且抑制作用存在剂量效应关系。结论:T3对高糖导致的胰岛细胞的氧化损伤有一定的保护作用,且存在浓度依赖性。

Objective To investigate the effect of 3, 5, 3'-Triiodothyronine (T3) on RIN-m5F cultured with high glucose. Methods RIN-m5F were cultured in RPMI-1640, and then divided into normal control group, high glucose group (glucose 44mM), α-lipoic acid group (200μM LA+ high glucose) , T3 groups (10-5M,10-7M,10-9M + high glucose) respectively. The cell viability, the level of intracellular ROS, MDA, SOD, T-AOC, insulin secretion and the mRNA expression level of MafA, Glut2, GCK were measured when cultured for 72h. Results Compare to control group, cell viability, SOD and T-AOC activity were significantly decreased , ROS generation and MDA content were significantly increased ,insulin secretion was significantly lower and the MafA , Glut2 , GK mRNA level were decreased in high glucose group.These changes were inhibited by LA and T3 with a dose-dependent manner, besides T3's effect on the mRNA level of GCK. Conclusion T3 protected RIN-m5F cell against injury induced by high glucose through inhibiting oxidation in dose-dependent manner.
