阴道非白念珠菌的PCR-RFLP鉴定
Identification of non-Candida albicans from Patients with Vovovaginal Candidasis by PCR-RFLP
目的 对分离自外阴阴道念珠菌病(VVC)患者阴道的常见致病性非白念珠菌进行PCR–RFLP鉴定。 方法 首先采用芽管试验、厚壁孢子试验、CHROMagar显色培养基及API试纸条将分离自VVC患者阴道内的念珠菌菌株鉴定到种,然后采用真菌通用引物将4种常见非白念珠菌(包括光滑念珠菌、近平滑念珠菌、克柔念珠菌和热带念珠菌)进行PCR扩增,并选用MspⅠ、HaeⅢ两种内切酶对扩增产物进行酶切分析。 结果 在4种非白念珠菌中光滑念珠菌15株(7.5%),近平滑念珠菌7株(3.5%),克柔念珠菌5株(2.5%),热带念珠菌2株(1.0%);PCR扩增后均产生稳定、清晰的条带,扩增产物经MspⅠ、HaeⅢ酶切后分别产生4种和2种特异性带型。 结论 外阴阴道念珠菌病非白念珠菌中以光滑念珠菌为主;PCR–RFLP可以快速、特异的鉴定非白念珠菌。
bstract: Objective To identify common pathogenic non-Candida albicans clinical isolated from patients with vulvovaginal candidasis by PCR-RFLP analysis method. Method Germ tube test,chlamydospore test,CHROMagar Candida and API20 kit system were applied to separate non-Candida albicans strains from Candida albicans.Then PCR was used to amplify the internal transcribed spacer region of ribosomal DNA from 4 species of common pathogenic non-Candida albicans(Candida glabrata,Candida parapsilosis,Candida krusei and Candida tropicalis),and the products were digested with the two restriction endonucleases(MspⅠ、HaeⅢ)respectively. Results The 4 isolates consisted of 170 strains of Candida albicans(85.0%), 15 strains of Candida glabrata(7.5%),7 strains of Candida parapsilosis(3.5%),5 strains of Candida krusei(2.5%),2 strains of Candida tropicalis(1.0%)Distinct bands of products with different molecular weights were obtained after amplification from 4 non-Candida albicans ,and the products were digested with MspⅠand HaeⅢ,producing 4 and 2 unique band patterns,respectively. Conclusion C.glabrata is the main pathogenic yeasts of non- Candida albicans in the vulvovaginal candidiasis.PCR-RFLP is rapid and specific for the identification of non- Candida albicans.
