益肾化浊法对慢性脑缺血血管性痴呆大鼠脑组织海马HIF-1α和VEGF表达的影响

目的:观察益肾化浊法对慢性脑缺血血管性痴呆大鼠脑组织海马HIF-1α和VEGF表达的影响。方法:采用双侧颈总动脉永久性阻断法建立血管性痴呆大鼠模型,造模后7d随机分为假手术组,模型组和聪圣颗粒低、中、高剂量组和尼麦角林片组,给药治疗60天后,采用Western-blot及Real Time-PCR方法检测大鼠海马HIF-1α、VEGF表达水平。结果:与假手术组比较,模型组大鼠HIF-1α和VEGF的蛋白表达量和mRNA的表达量均升高(P<0.05);与模型组比较,聪圣颗粒中、高剂量组可升高模型组HIF-1α及VEGF的蛋白和mRNA的表达(P<0.05,P<0.01)。结论:益肾化浊法对慢性脑缺血血管性痴呆大鼠作用机制可能与调控海马区HIF-1α和VEGF的表达有关。
Objective:To observe the expression of HIF-1α and VEGF in The hippocampus of brain tissue with the Chronic ischemic vascular dementia illness treating with the method of tonifying kidney and resolving turbidit. Methods: The bilateral carotid artery of rats were occluded to establish the model of permanent vascular dementia.7 days after the operation ,they were randomly divided into 6 groups:sham ,model, low, medium and high dosage group of Congsheng Granules and Nicergoline Tablets group. After intervention of 60 days, we begin to detectthe HIF-1αand VEGF expression in hippocampus area of rats by means of Western-blot and Real Time-PCR. Results:Compared with the sham group, the expression levels of HIF-1α and VEGF protein and mRNA of model group were increased significantly (P<0.05); compared with the model group, middle and high dosage group of Conheng Granules incrasethe expression of HIF-1αand VEGF protein and mRNA (P<0.05, P<0.01). Conclusion: The method of tonifying kidney and resolving turbidit may regulate the expression of HIF-1αand VEGF in hippocampus of rats to protect the brain against Chronic ischemic vascular dementia.?????
