芦荟大黄素抑制LPS诱导RAW264.7细胞释放炎症因子作用机制研究

目的:以LPS诱导的RAW264.7细胞炎症模型,探讨芦荟大黄素的抗炎作用机制。方法:以5μΜ、10μΜ、20μΜ三个浓度的芦荟大黄素预处理RAW264.7细胞1h后,给予1μg/mL的LPS诱导24h。以ELISA法检测IL-1β、IL-6的生成量,以qRT-PCR方法检测IL-1β、IL-6 mRNA表达。结果:芦荟大黄素中、高剂量组可显著抑制IL-1β、IL-6的生成量(P<0.05,P<0.001),IL-1β、IL-6 mRNA的表达(P<0.05,P<0.001),且均随浓度增加抑制作用增强。结论:芦荟大黄素具有较好的抗炎活性,可抑制炎症细胞因子IL-1β、IL-6的释放,该活性通过抑制它们的转录表达发挥的作用。
Objective: To explore the anti-inflammatory mechanism of aloe-emodin in LPS-induced RAW264.7 cells. Methods: RAW264.7 cells treated with 1μg/mL of LPS for 24h after pretreated with aloe-emodin for 1h which is three concentrations (5μΜ, 10μΜ, 20μΜ). The amount of IL-1βand IL-6 was detected by ELISA assay. The expression of IL-1βand IL-6 mRNA was detected by qRT-PCR. Results: Aloe-emodin significantly inhibited the release of IL-1βand IL-6 (P <0.05, P<0.001), the expression of IL-1βand IL-6 mRNA (P<0.05, P<0.001) in a dose-dependent manner. Conclusion: Aloe-emodin has excellent anti-inflammatory activity, and the mechanism depends on inhibit the transcriptional activity of pro-inflammatory cytokines.
