苏云金芽胞杆菌dlt操纵子功能研究
Study on the biofunction of dlt operon in Bacillus thuringiensis
dlt操纵子在芽胞杆菌中广泛存在,主要由dltA、dltB、dltC、dltD四个结构基因组成,其编码的蛋白质能修饰细胞壁的主要成分磷壁酸发生烷基化,使细菌细胞壁表面生成净正电荷,从而使芽胞杆菌对阳离子抗菌肽的抵抗力增强。本研究构建通过同源双交换构建了苏云金芽胞杆菌无晶体突变株BMB171菌株dlt操纵子突变株,发现突变株生物量显著降低,且细胞表面负电荷增加。结果表面dlt操纵子不仅能决定苏云金芽胞杆菌细胞表面电荷,而且影响细胞的生长与繁殖,预示着该操纵子对苏云金杆菌具有非常重要的生物学意义。?????
lt operon are widespread in Bacillus, mainly composed of four structural genes dltA, dltB, dltC and dltD ,and its encoded protein can modify cell wall of the main components of the teichoic acid alkylation, formed on the surface of the bacterial cell wall, make a net positive charge, so that the bacillus of cationic antimicrobial peptide resistance increased. This study attempts to construct Bacillus thuringiensis dlt operon mutant, and the characters of the control cell surface charge were determined. By homologous recombination technology knockout of Bacillus thuringiensis acrystalliferous mutant strain BMB171 the dlt operon, compare the difference of the parent strain and the mutant strain growth, cell wall surface charge differences. This study successfully constructed the BMB171 dlt operon mutant, and the traits were determined. To verify the dlt operon also can make the surface of Bacillus thuringiensis net positive charge is increased, which laid a good foundation for subsequent further research of Bacillus thuringiensis dlt operon of biological function, in order to improve the Bacillus thuringiensis insecticidal toxicity to find new ideas.
