雌二醇对经历强迫游泳应激大鼠中缝核色氨酸羟化酶与5-羟色胺含量的影响

目的 观察雌二醇对经历强迫游泳应激大鼠中缝核色氨酸羟化酶(TPH)与5-羟色胺(5-HT)含量的影响,探讨雌激素与应激在女性抑郁症发病机制中的作用。 方法 手术切除35只成年雌性SD大鼠双侧卵巢后3周,随机分成5组(n=7):非应激组、对照组、雌二醇组、氟西汀组、雌二醇+氟西汀组。各组慢性给药14 d,第14 d,非应激组大鼠除外,其他四组进行15 min强迫游泳实验(FST),FST结束后2 h,各组大鼠经4%多聚甲醛溶液灌注固定、取脑,制备冰冻脑组织切片,通过间接免疫荧光染色法观察大鼠中缝核TPH和5-HT的含量。 结果 (1) 对照组大鼠中缝核TPH与5-HT阳性区域的积分光密度值(IOD)显著低于非应激组(P<0.01)。(2)雌二醇、氟西汀和雌二醇+氟西汀组大鼠中缝核TPH和5-HT阳性区域IOD值均显著高于对照组(P<0.05)。 结论 强迫游泳应激可导致大鼠中缝核TPH和5-HT含量减少,而预先给予雌二醇可阻止这种变化,与抗抑郁药物氟西汀作用相似。这为理解雌激素与应激在女性抑郁症发病机制中的作用提供了实验依据。
Objective To investigate the effect of estradiol (E2) on tryptophan hydroxylase (TPH) and 5-hydroxytryptamine (5-HT) content in raphe nuclei of rats subjected to forced swimming stress, explore the role of estrogen and stress in disease mechanism of depression in women. Methods Three weeks after the ovariectomy, 35 ovariectomized (OVX) female SD rats were randomly divided into 5 groups (n=7): the non-stress group, the control group, the estradiol (E2) group, the fluoxetine (FLX) group and the E2+FLX group. Animals were administered with different drugs for 2 weeks. On the fourteenth day, animals except those in the non-stress group were subjected to the 15-min forced swimming test (FST). 2 hours after the FST, all animals including those in the non-stress group were perfused with 4% paraformaldehyde and brains were removed for TPH and 5-HT immunofluorescence staining. We compared the content of TPH and 5-HT by observing and calculating the integrated optical density (IOD) of immunofluorescent-positive signals in raphe nuclei. Results (1) The IOD value of TPH- and 5-HT-positive region in raphe nuclei of rats in the control group was significantly lower than that of non-stress group (P<0.01). (2) It has been shown that the IOD value of TPH- and 5-HT-positive region in raphe nuclei of rats in the E2 group, FLX group and E2+FLX group was significantly higher than that in the control group (P<0.05). Conclusion Forced swimming stress can decrease the TPH and 5-HT content in raphe nuclei. Such changes can be prevented by giving estradiol in advance. Same results were observed with antidepressant-fluoxetine. These effects may underlie the role of estradiol and stress in the disease mechanism of depression in women.
