本文采用半定量RT-PCR的方法,初步研究了可作为外植体的大蕉 (Musa ABB,cv. Dajiao)各组织及相关组织培养物中SERK (SOMATIC EMBRYOGENESIS RECEPTOR KINASE)基因的表达模式, 试图确定SERK基因的表达与香蕉组织培养物胚性形成的关系。研究结果表明,在大蕉的根、假茎、叶、果实、果皮和花序中都能检测到SERK基因的表达,但SERK基因在花序的表达量明显高于其他组织器官。这与目前认为花序是用于胚性愈伤组织诱导的理想外植体的研究结论相吻合。大蕉薄片培养所得的三种愈伤组织中, 其中一种经继代可从中得到的胚性愈伤组织(称类型II愈伤组织),SERK 也在这类愈伤组织中表达最强。因此,可将SERK的表达作为快速检测香蕉培养细胞胚性高低的指标,用于优化诱导香蕉胚性愈伤组织的培养条件。

o understand the relationship between the expression of the SERK-like gene and the embryogenic competence of banana (Musa ABB,cv. Dajiao) in vitro cultures. In this study the semi-quantitative RT-PCR method was used to analyse the expression of SERK gene in the different explants which may be used in tissue culture. The explants included pseudo stem, roots, leaves, immature male flower and young fruit as well as callli induced from micro-cross section of the pseudo stem cultured in three different culture system. The results showed that a certain level expression of the SERK gene could be found in the tested roots、fruit、pseudo stem and immature flowers,however the expression level of this gene in the immature flowers was higher than that in the other tested tissues. It could be a good explanation for why the immature flowers are the best explants for embryogenic callus induction of banana in many reported papers elsewhere. Analysis of expression of the SERK gene in three calli, induced from the micro-cross section on three media, revealed that the expression level of type II callus which was the embryogenic callus was higher than that in other two kind calli. Therefore, there is closed correlation between the expression of the SERK gene and the embryogenic competence of banana callus , and the SERK gene is proved to be an efficient molecular marker for optimizing the culture condition to obtain the embryogenic cultures of banana , at least for Musa ABB,cv. Dajiao in micro-cross section culture system.
