目的:通过体外细胞实验,探究抗肿瘤药物托瑞米芬(TOR)对血管平滑肌细胞(VSMCs)的细胞生物学行为的影响及其作用机制,旨在为TOR能否应用于药物洗脱支架提供一定的理论依据。方法:运用MTT法,流式细胞术法,划痕法,免疫荧光和免疫印记的方法探讨TOR对平滑肌细胞的影响。结果:该药物能抑制平滑肌细胞增殖;促进平滑肌细胞凋亡;显著抑制平滑肌细胞群体迁移行为;随着TOR浓度增高,PCNA表达量明显降低;P53的表达随TOR浓度的增加而增加;TOR显著抑制了Integrin β1及Rho的表达,降低了ROCK信号通路中下游因子MLC和pMLC的表达水平。结论:TOR抑制细胞的增殖和迁移,促进细胞的凋亡,并通过Integrin β1- Rho- ROCK- (p)MLC信号通路抑制细胞的迁移行为。?????

Objective: In this study, using vascular smooth muscle cells as cell model in vitro, aiming to investigate the effect of toremifene (TOR), a new generation of antineoplastic drugs, on biology behavior and migration mechanism of vascular smooth muscle cells (VSMCs), the effect of toremifene against restenosis (RS) was investigated. Methods: MTT method, flow cytometry method and scratch wound assay were used to examine cell growth, apoptosis and migration. Immunocytochemistry and immunoblotting were used to examine the expression of PCNA, P53 and protein of Rho/ROCK signal transmission pathway. Results: The high concentration of TOR can inhibit cell proliferation and the inhibiting effect was gradually enhanced with increasing TOR concentration; TOR can significantly promote cell apoptosis; TOR can significantly inhibit the group cell movement and migration behavior; the higher concentration of TOR treatment inhibited cell growth and induced cell apoptosis, the expression of PCNA decreased with the increasing TOR concentration; the expression of P53 increased with the increasing TOR concentration, which indicates that TOR accelerated cell apoptosis by promoting expression of apoptosis gene P53; TOR significantly inhibited the expression of Integrin β1 and Rho, and decreased the ROCK signaling pathways factor MLC and pMLC expression level. Conclusion:Our results indicated that TOR may inhibit cell growth and induce cell apoptosis and inhibit cell migration behavior by the Integrin β1- Rho- ROCK- (p) MLC signaling pathway.??????????
