目的:探讨力学刺激对体外培养口腔鳞状细胞癌KB细胞的凋亡基因caspase-3表达的变化。方法:采用FX-4000柔性基底加载系统对人口腔鳞状细胞癌KB细胞进行体外加载培养,卸载后运用流式细胞仪检测分析KB细胞的凋亡率,通过实时荧光定量PCR检测分析凋亡基因caspase-3表达的变化。结果:在加载程式(方波,幅度15%,频率0.5Hz,时间24h)的拉伸培养下,KB细胞的caspase-3 mRNA水平高于正常对照组,差异具有统计学意义(P<0.05)。结论:力学刺激能够诱导凋亡信号通路中的caspase家族产生级联反应,通过激活caspase-3因子而诱导人口腔鳞状细胞癌KB细胞发生凋亡。

Objective:To study the effects of Mechanical stimulation on caspase-3 protein expression of Human oral Squamous Cell Carcinoma KB Cells in vitro. Method: The Human oral Squamous Cell Carcinoma KB Cells was cultured by FX-4000 Strain unit, The apoptotic rate was detected by flow cytometry, The protein expression of caspase-3 was examined by Real-time fluorogenic quantitative polymerase chain reaction (FQ-PCR). Result: The protein expression of caspase-3 increased significantly compared with control after the KB cells were subjected to regimens(square wave,15% elongation amplitude,0. 5Hz,24h ) , the difference had statistical significance( P < 0. 05). Conclusion: Mechanical stimulation activated caspase-3 which induced the KB cell apoptosis, therefore caspase-3 is involved in the mechanical stimulation-induced cell apoptosis.
