本论文通过比较3-((2,4-二甲氧基苯基)(羟基)亚甲基)-1-异丙基吡咯烷-2,4-二酮的4种不同合成方法和研究,找到一条以2,4-二羟基苯甲酸作为起始原料合成2,4-二羟基苯甲酸酯,然后在碱作用下与乙酸乙酯反应缩合制备2,4-二甲氧基苯甲酰基乙酸乙酯,进而与相应的2-异丙基氨基乙酸乙酯缩合制得目标化合物,具有原料价格低廉、操作简单、易于控制、收率稳定的特点,为高活性化合物3-((2,4-二甲氧基苯基)(羟基)亚甲基)-1-异丙基吡咯烷-2,4-二酮的进一步应用奠定了基础。

In this paper, through comparison of the four routes for synthesis of 3-((2,4-dimethoxy phenyl)(hydroxy)methylene)-1-isopropyl-pyrrolidine-2,4-dione, 2,4-dimethoxybenzoic acid was selected as starting material. Firstly, it was transformed into 2,4-dimethoxybenzoate and then, at the presence of base, 2,4-dimethoxybenzoate was transformed into 2,4-dimethoxybenzoylacetate. Finally, 3-((2,4-dimethoxyphenyl) (hydroxy)methylene)-1-isopropyl-pyrrolidine-2,4-dione was prepared by reaction of 2,4-dimethoxybenzoylacetate with 2-isopropylaminoacetate. This procedure possess these features: cheap materials, operations simple, easy to control, stable yield. This work was also helpful to further application of the target compound.
