活血利水法对兔外伤性玻璃体视网膜病变增殖膜中转化生长因子信使核糖核酸表达的影响

目的:探讨活血利水之散血明目片对外伤性增生性玻璃体视网膜病变(tPVR)增殖膜(ERM)中EGFmRNA表达的影响。方法:将40只成年有色家兔随机抽取32只,造成外伤性PVR模型,随机分成活血利水组、活血化瘀组、利水明目组、模型组,另8只为空白组。连续灌胃30d后,观察眼底PVR分级情况,免疫组织化学方法检测中EGFmRNA表达在各组的表现及病理组织学改变。结果:在活血利水组,ERM中EGFmRNA阳性程度低于模型组,差异具有显著性(P<0.01)。活血化瘀组与利水明目组也能降低中EGFmRNA表达的阳性程度,与模型组相比有显著性意义(P<0.05)。活血利水组中EGFmRNA阳性程度低于另二个治疗组,有显著性差异(P<0.05)。活血利水治疗组中EGFmRNA阳性程度略高于空白组,有显著差异性(P<0.01)。结论:活血利水法是活血化瘀和利水明目两者作用的协同,能通过拮抗增殖膜中EGFmRNA表达的作用来抑制增殖细胞的过度增生,从而防治PVR形成和发展。
Objective: To study the curative and preventive effects and mechanism of HuoXueLiShui Method (HXLSM) on the effects of EGFmRNA’s expression on ERM of Traumatic Proliferative Vitreoretinopathy. Methods: 32 rabbits were selected randomly from 40 healthy adult color rabbits and induced to traumatic proliferative vitreoretinopathy models, then divided into four groups: the first group was treated with San Xue Ming Mu Tablet-Huo Xue Li Shui (HXLS) group, the second treated with Zhi Xue Qu Yu Tablet-Huo Xue Hua Yu (HXHY) group, the third treated with Zhu Ling San-Li Shui Ming Mu (LSMM) group and the model group. The rest 8 formed the blank control group.After the rabbits had been fed and administrated for a month, we observed the visibility of fundus, the degree of PVR and pathologic changes, measured EGFmRNA in epiretinal membrane(ERM) expression were observed with immunohistochemistry technique. Results: Compared with model group, the positive level of EGFmRNA, was decreased significantly in HXLS group (p<0.01). Compared with model group, the positive level of EGFmRNA decreased much in HXHY and LSMM group(p<0.05). And the level of EGFmRNA in HXLS group were lower than those in HXHY and LSMM group. The positive level of EGFmRNA in HXLS group was a little higher than those in blank group (p<0.05). The PVR in HXLS group was lighter than that in model group、HXHY and LSMM group. Conclusions: Huo Xue Hua Yu Method is the coordination of Huo Xue Hua Yu method and Shui Ming Mu method. It could inhibit and cure the formation and development of traumatic PVR by counteracting the expression of EGFmRNA on ECM as to inhibiting proliferrative cell’s hyperplasy.
