目的:探讨地塞米松是否能够通过调节PPARγ表达影响滋养层细胞的迁移功能。方法:培养人绒毛膜癌细胞株JEG-3,Real-time PCR检测地塞米松对PPARγ的mRNA表达调控,western blot检测地塞米松对PPARγ蛋白表达的影响。Transwell试验检测地塞米松和过表达PPARγ对滋养层细胞迁移功能的影响。结果:地塞米松可以使人滋养层细胞PPARγ表达下调。过表达PPAR-r可抑制滋养层迁移,地塞米松促进滋养层细胞迁移。结论:地塞米松可以下调PPARγ的表达,影响滋养层细胞的迁移功能,这也许可以解释PPARγ对胎盘发生的重要作用。

o explore whether dexamethasone can modulate trophoblast migration via regulating the expression of PPARγ. METHODS: The regulation of dexamethasone on PPARγ in cultured human choriocarcinoma cell line JEG-3 was analysed by real-time PCR and western blot.The Transwell analysis is taken for migration of dexamethasone or over-expression of PPARγ treated and control cells . Results: Dexamethasone inhibits PPARγexpression. PPARγoverexpression inhibits trophoblast migration while dexamethasone promote cell migration in JEG 3. Conclusion: Dexamethasone promotes the migration of trophoblast by down-regulating the expression of PPARγ, which may partly explain the important role of PPARγand dexamethasone in human placenta.
