青光安对兔眼滤过性术后中I型胶原蛋白的影响

bstract:Object: To observe effects of four effective components of Qing Guang An on typeⅠcollagen of scarring tissue of filtration canal after glaucoma surgery, and discuss their effect on preventing its formation, and a preliminary judgment on their toxic effects.Methods: 42 healthy adult New Zealand long ears white rabbit, were divided into 7 groups randomly with 6 rabbits in each group. These seven groups were used as following: group A was blank group;group B was model group; group C was a control group of MMC; group D, E, F, G was used effective component signed respectively as 1, 2, 3, 4. The next day after molding began to use medicine on each group. after surgery, the formation of filtering bleb was observed , and the intertion status and eyes' general condition were checked. After using medicine for four weeks，After dyed by HE organization structure where the surgery had done. Expression of typeⅠcollagen in Tenon bourse in filtering bleb area was checked with the immunohistochemical method.Result: C, E groups remained effective filtering bleb. Filtering bleb in group B turned into no function or disappeared two weeks after surgery. Group D, F, G had part of filtering bleb existing four weeks after surgery. Comparing with the the expression of typeⅠcollagen, except group C and E, B and G, F and G, result of others have obvious significance on statistics showing as P<0.05. According to the observation of common condition after surgery and specimen polarizing at the fourth week, the group with MMC had complication after surgery, other groups with effective components did not show any abnormal lesions in eye organization structure.Conclusion: (1) In doing conventional trabeculectomy and iridodialysis, a filtration surgery model will be successfully built. (2) The effective componment 2 of Qing Guang An and MMC can obviously inhibit the expression of typeⅠcollagen,reduce the hyperplasia of scar tissue,which has obvious effect for preventing scarring of filtration canal. (3) It suggests that orally taking the effective componment 2 of Qing Guang An does not has obvious side effects, which is much more safe than MMC, and may be a new and safe medicine to preventing scarring of filtration canal after glaucoma surgery and thus improve the success rate of the surgery.
