In order to study the usefulness of low-molecular-weight glutenin subunits (LMW-GS) to wheat quality improvement, we characterized six LMW-GS genes, which were titled 1-50,2-11,2-34,1-31,1-36,1-95, from an introgression line II-12 between Triticum aestivum cv. Jinan177 (JN177) and Agropyron elongatum using genomic PCR method. All six sequences have high similar to those of wheat and related grasses reported before. Analysis of their deduced amino acid sequences suggests that 1-50,2-11,2-34 may be real genes and the other three are pseudogenes according to whether or not containing in-frame stop codon. Subunit 1-50 contains nine Cys residues and three may be involved in inter-molecular disulfide bonds; Glutamine in mature subunit 2-34, occupying about 34.29% of deduced amino acid, is significantly higher than the mean value of LMW-GS in common wheat. Thus, the two genes may be responsible for high quality of the hybrid line II-12.
