尿素包合富集羊毛脂中支链脂肪酸的研究

Enrichment of branched chain fatty acids from lanolin via urea complexation

BCFA作为常见食品中的生物活性成分,但关于其富集的报道却微乎其微。本研究以羊毛脂为原料制取羊毛酸,对羊毛脂脂肪酸组成进行分析;采用尿素包合法对羊毛脂中的BCFA进行富集,以支链脂肪酸含量和回收率为考察指标,通过单因素实验优化工艺条件。尿素包合富集羊毛脂中BCFA的最佳工艺条件:结晶溶剂为95%乙醇、尿素与脂肪酸质量比为2:1、尿素与95%乙醇质量体积比为1:6、结晶温度为4℃、结晶时间为12h。在此条件下BCFA含量从64.35%提高到94.69%,回收率为40.02%。此外母乳中主要的六种BCFAiso-C14:0、iso-C15:0、anteiso-C15:0、iso-C16:0、iso-C17:0、anteiso-C17:0,含量达到75%以上。

lthough branched-chain fatty acids (BCFA) are bioactive components of common food fats, little research has been performed on BCFA-enrichment technologies. In this study, lanolin acids were prepared from lanolin and its fatty acid composition was analyzed. The BCFA in lanolin were enriched by urea complexation. With content and recovery of BCFA as the indexes, the enrichment conditions were optimized by single factor experiment. Under optimal conditions (95% aqueous ethanol/urea ratio: 6, urea/FA ratio: 2, crystallization temperature: 4 C, crystallization time: 12 h), the content and recovery yield of BCFA from lanolin were obtained as 94.69% and 40.02%, respectively. In addition, the six major BCFAs of breast milk were found to be iso-C14:0, iso-C15:0, anteiso-C15:0, iso-C16:0, iso-C17:0, and anteiso-C17:0, whose contents in breast milk were more than 75%.
