乌饭树叶多酚氧化酶参与黑色素形成及其酶学性质的初步研究
Study on polyphenol oxidase from Vaccinium Bracteatum thunb leaves involved in the formation of melanin and some enzymatic properties
以乌饭树叶为原料,制备乌饭多酚氧化酶(PPO)粗酶液,研究乌饭树叶本身的PPO是否参与乌饭树叶黑色素的形成;在粗酶液的基础上,采用硫酸铵盐析、透析对酶进行初步纯化,并对其部分酶学性质进行分析。结果表明:乌饭树叶中的PPO参与了乌饭树叶黑色素的形成;纯化所得的酶活为:112.9 U/mg,纯化倍数:2.2,酶活回收率为:25.0%;该酶的最适pH为6.5,最适温度为45℃左右,最适底物为邻苯二酚。本实验为乌饭树叶黑色素的制备提供了一定的参考依据。
he crude polyphenol oxidase(PPO) was extracted from leaves of Vaccinium Bracteatum Thunb. First of all, experiments were performed to prove that PPO was involved in the formation of melanin. Then, the crude PPO was preliminary purified by ammonium sulfate precipitation method and some enzymatic properties of PPO was studied. The results showed that the specific activity of the purified PPO was 112.9 U/mg, which exhibited a purification fold of 2.2and a yield of 25.0%. The optimal pH and temperature for PPO were 6.5 and 45℃, respectively. The optimal substrate is catechol. This could provide some reference basis for the preparation and application of melanin from Vaccinium Bracteatum Thunb.
