【目的】本文针对马铃薯Y病毒的功能基因进行扩增、克隆和生物信息学分析。【方法】首先提取病株的RNA进行单基因克隆,获得8个功能基因的测序结果,然后将基因序列翻译成氨基酸序列,综合使用多种生物信息学方法分析氨基酸序列。【结果】根据TMHMM预测P3蛋白后半部分有一个跨膜螺旋序列;SignalP分析结果显示8个蛋白的N端均无信号肽,均属于非分泌型蛋白;COILS Server 预测可知P1、HC-Pro、P3、CI、VPg、NIb包含卷曲螺旋区域;使用PSORT II Prediction 工具进行亚细胞定位,结果证明这些蛋白有多个定位位置。【结论】成功完成了8个功能基因的生物信息学分析,为PVY功能基因的功能研究和抗体的制备提供参考价值。

【Object】In this paper, we have studied the eight important genes in potato virus Y with gene amplification and bioinformatic analysis. 。【Method】First, we extract RNA from potato leaves exhibiting typical symptoms and clone single gene, then translate these gene sequences into amino acid sequences,the last we import these sequences into related bioinfomatic software to gain the physicochemical properties and the parameters of the structure domains and the subcellular localization. We found three stable proteins (HC-Pro, CI and NIa) and five instable proteins (P1, P3, VPg, NIb and CP). According to the GRAVY, we can know these proteins can dissolved in water. We found a transmembrane in the latter half of P3 protein according to the TMHMM, at the same time we found no any signal peptide in the eight proteins according to the analysis of SignalP, hence there are no secretory proteins in these functional proteins. P1, HC, P3, CI, VPg and Nib have coil regions on the basis of the COILS Server analysis. The subcellular location by PSORT II Prediction suggest these proteins have more than one locations.
