转录因子ATF6调控下游基因CTH表达并影响肝细胞癌进展
Regulation of transcription factor ATF6 on the expression of CTH and the progression of Hepatocellular carcinoma
目的:探究转录因子ATF6对肝细胞癌HCC的调控作用,阐释ATF6对HCC细胞发展与转移的影响并验证ATF6对下游基因CTH基因表达的调控作用,验证CTH调控HCC细胞中H2S的生成。方法:在HCC细胞系SMMC-7721细胞中瞬时转染ATF6过表达及敲低质粒并通过transwell迁移与侵袭、集落形成实验研究ATF6对于SMMC-7721肝癌细胞的影响,通过Western Blot及qPCR实验验证CTH是ATF6的下游靶基因,瞬时转染CTH过表达质粒并利用亚甲基蓝方法检测细胞中H2S的生成。结果:过表达ATF6的SMMC-7721细胞集落形成、迁移与侵袭现象明显增多,细胞中CTH的mRNA及蛋白表达水平显著上升,CTH过表达的SMMC-7721细胞中H2S生成明显增多。结论: ATF6促进SMMC-7721肝癌细胞的侵袭、迁移和生长,并提高下游基因CTH的表达水平,CTH促进SMMC-7721细胞中H2S的生成,ATF6参与调控下游基因CTH的表达并影响肝细胞癌的进展。
Objective: Explore the regulatory effect of transcription factor ATF6 on Hepatocellular carcinoma(HCC)and explain the influence of ATF6 on development and metastasis of HCC cells, verify the regulatory effect of ATF6 on the expression of the CTH gene, examine the production of H2S by CTH in HCC cells. Methods: Transfect HCC cells with ATF6 overexpression or knockdown plasmids and detect the effect of ATF6 on HCC cells through transwell migration, invasion and clonogenic survival assays, Western Blot and qPCR experiments were conducted to verify that ATF6 regulate the expression of CTH, H2S production was detected by Methyl blue method in CTH overexpressed HCC cells. Results: The migration and invasion of ATF6-overexpressed HCC cells increased significantly, ATF6 markedly increased the expression level of CTH mRNA and protein, H2S production in CTH-overexpressed HCC cells was significantly improved. Conclusion: ATF6 promotes the invasion, migration and growth of HCC cells, and improves the expression level of downstream gene CTH, CTH promotes the production of H2S in HCC cells, and ATF6 is involved in regulating the expression of CTH and affecting the progress of hepatocellular carcinoma.
