【目标】植物乳杆菌ZS2058(ZS2058)和丁香酚均能不同程度地保护沙门氏菌感染小鼠,本研究旨在探究ZS2058联合丁香酚对小鼠沙门氏菌感染的预防作用及其机制。【方法】本研究通过96孔板-肉汤稀释法检测丁香酚的抑菌活性;通过HT-29细胞模型研究ZS2058和丁香酚对沙门氏菌粘附侵袭能力及其毒力基因表达的影响。在动物实验中,小鼠在感染沙门氏菌前10天,分别口服PBS,ZS2058,丁香酚,ZS2058和丁香酚,观察并记录感染后20天小鼠存活情况,对小鼠肠道菌群进行16S rDNA测序分析,通过ELISA试剂盒检测小鼠肝脏炎症水平的变化。【结果】存活率(SR)结果表明,ZS2058联合丁香酚(CLPZE)(SR= 80%)对沙门氏菌感染小鼠(SR = 60%)的预防效果是ZS2058(SR = 70%)的2倍,是丁香酚(SR = 63.3%)的6倍。ZS2058和丁香酚能协同抑制沙门氏菌生长,降低沙门氏菌粘附侵袭能力及其毒力基因表达;维持沙门氏菌感染小鼠肠道稳态,增强小鼠免疫反应以及时清除病原体。【结论】ZS2058联合丁香酚在预防小鼠沙门氏菌感染中存在协同作用。

[Aim] Previous studies have reported the preventive effects of Lactobacillus plantarum ZS2058 (ZS2058) and eugenol against Salmonella in murine models. This work aims to study the preventive effect of ZS2058combined with eugenol against Salmonella infection in mice and the involved mechanisms. [Methods] Bactericidal activity was evaluated by micro-broth dilution assays. HT-29 cells model and PCR were used to investigate the effects of ZS2058 and eugenol on the invasiveness and virulence gene expression of Salmonella. The synergistic effect was assessed by coefficient drug interaction (CDI). C57BL/6 mice (female, 8 wk old) were pretreated with PBS (group M), ZS2058 (group Z), or eugenol (group E), or ZS2058 and eugenol (group ZE) for 10 days prior to infection with Salmonella, the mortality rate was recorded daily for 20 d. The intestinal microbiota of cecum contents was characterized by 16S rDNA Gene Sequencing. The cytokines in liver were detected by ELISA. [Results] ZS2058 combined with eugenol (CLPZE) treatment (SRZE, 80%) increased the survival rate (SR) of ST-infected mice (SRM, 60%) to two-fold compared with that of treated by ZS2058 alone (SRZ, 70%), and six-fold compared with the effect of eugenol alone (SRE, 63.3%). CLPZE had a synergistic effect on inhibiting ST growth, reducing its invasiveness and downregulating virulence gene expression in vitro. CLPZE helped the host form a healthier gut ecosystem,and elicited a stronger and earlier immune response against systemic infection. [Conclusions] Our studies provide substantial evidence strongly suggesting that CLPZE has a synergistic effect in preventing Salmonella infection in C57BL/6 mice.
