34+细胞体外红系分化模型的建立及ZNF268的表达检测

研究表明ZNF268基因可能与红系造血发育相关。为了确定ZNF268基因在红系分化中的表达情况,本文利用免疫磁珠吸附等方法,获得纯化的CD34+造血干/祖细胞,利用细胞因子联合诱导其向红系分化;在诱导的不同阶段,利用流式细胞术和瑞氏吉姆萨染色,检测细胞表面分子的表达和细胞形态的变化,并检测在此过程中ZNF268蛋白的表达情况。结果显示,我们成功构建了CD34+细胞向红系方向的体外诱导分化模型,并检测到ZNF268的两种蛋白产物ZNF268a和b2在红系分化过程中表达均下调,这些结果暗示ZNF268可能在红细胞的发育过程中发挥作用。
Previous studies showed that ZNF268 gene might be involved in the erythroid hematopoietic development. To determine the expression of ZNF268 gene during erythropoiesis differentiation, CD34+ hematopoietic stem/progenitor cells be isolated and purified using immunomagnetic beads adsorption methods, and then be induced into erythropoiesis differentiation by combined-cytokines-induced methods. At different inducing stages, the cell surface molecular marks and changes in cell morphology be checked by flow cytometry and Wright's Giemsa staining, and ZNF268 expression in this process be detected. The results show that the cytokines-inducted erythroid differentiation of CD34+ cells in vitro be successfully developed and the the expression of ZNF268a and b2 encoded by ZNF268 gene decreased significantly during erythroid differentiation, and the results suggest that ZNF268 maybe play a role during the erythroid hematopoietic development.
