目的 检测转化生长因子β1(transforming growth factor, TGF-β1)能否在体外诱导大鼠肝卵圆细胞上皮-间质细胞转变(epithelial-mesenchymal transition,EMT)及其相应的信号转导机制。方法 在体外培养的大鼠肝卵圆细胞(WB F344))中加入不同剂量的TGF-β1后,于48h后观察细胞形态的变化并收取细胞,分别采用Real-time PCR及Western blot检测上皮及间质细胞标志物表达的改变。结果 与TGF-β1未处理组相比,加入TGF-β1到WBF344细胞后,形态学上,TGF-β1诱导WB F344细胞向成肌纤维细胞样细胞转变,并且其间质细胞标志物表达上调,上皮细胞标志物的表达下调。随着TGF-β1剂量的加大,间质细胞标志物上调明显,上皮细胞标志物下调也明显(p<0.05)结论TGF-β1能在体外诱导大鼠肝卵圆细胞向间质细胞转变。

Purpose The aim of the present study was to investigate whether or not transforming growth factor-β1(TGF-β1)could induce epithelial-mesenchymal transition(EMT)of rat hepatic oval cells (WB F344)in vitro and its possible mechanism. Methods After the in vitro cultured WBF344 cells were treated with TGF-β1 at different doses.expression of the markers of epithelial cell and mesenchymal cell was assayed using Western blot and Real-time PCR analysis. Morphological alterations were examined by phase-contrast microscope. Results Incubation of WB F344 cells with TGF-β1 resulted in the up-regulation of the expression of the mesenchymocyte markers and the down regulation of the epithelial markers. With the increasing doses of TGF-β1, there were significantly higher expression of the mesenchymocyte markers and lower expression of the epithelial markers. Meanwhile,incubation of WBF344cells with TGF-βI could also lead to the transition of cells to myofibroblast-like cells morphologically.Conclusions TGF-β1 can induce EMT of WBF344 cells in vitro
